Height: 149 cm, Weight: 37 kg. Born in November 1863 (Bunkyū 3). Sagittarius, blood type B. First appeared at age 16. Hobby: Searching for Aoshi-sama. Refers to herself as "atashi" (sometimes "watashi").
A lively girl in love with Aoshi. Tomboyish, selfish, and emotionally expressive. Granddaughter of the former leader of the Oniwabanshū (Aoshi's mentor). Meets Kenshin while heading to Kyoto on the Tōkaidō and accompanies him there. At that time, she had lost her travel money and was robbing villains. She masters the Oniwabanshū-style kunai techniques and the Oniwabanshū-style martial arts passed down from Hannya, boasting skills beyond ordinary people. However, compared to Kenshin and others, her abilities are significantly lower, so she is not considered a combat asset, resulting in fewer battle scenes.
She is also fond of Aoshi's subordinates (Oniwabanshū), especially calling Hannya with the honorific "kun." She calls Shikijō with "san," but calls Hyottoko and Beshimi without honorifics, indicating she doesn't evaluate them highly. Due to her petite and childlike appearance, she is often seen as younger than her actual age (which she is somewhat concerned about). Saitō and Sanosuke call her "weasel girl" (since Kaoru is associated with a tanuki and Megumi with a fox), but she dislikes it (in the first anime series, Sanosuke often calls her this).
In the Kyoto arc, after Aoshi and Okina fought, she declared herself the leader of the Oniwabanshū and showed a willingness to oppose Aoshi. After Aoshi returned to Aoiya, she no longer called herself the leader, but in the first anime series, she continues to do so after the Kyoto arc.
In the Jinchu arc, she often teams up with Yahiko, using her energy to gather information. She barges into the police station in an obvious disguise to bring Saitō out, causing the group to be unimpressed. In the final stages, she destroys the "barrier" created by Kuroboshi's sea mines, paving the way for the final battle.
During the "Spring Cherry Blossoms" period, she was apparently undergoing bridal training. At this point, she is 21 years old, but her personality remains unchanged, and her appearance hasn't changed much either. In "Ashitaro the Ex-Con," she is seen near the five-story pagoda with Aoshi in the final scene.
In the first anime series, during the Shimabara arc, she sees the pattern on the body of the first victim of Shougo Amakusa and warns Kenshin and the Oniwabanshū members of the danger. She later finds Shougo but is defeated and lets him escape, then accompanies Kenshin to Shimabara. After the incident is resolved, she appears in a dress and returns with Kenshin and the others. In the Black Knights arc, she tracks the movements of the Sanada ninja army, helps Kenshin with Kuroi and Shiroi, and joins the search for the elixir. She then temporarily stays in Tokyo but returns to Kyoto with Aoshi and others.
In the PSP game "Re-Sen," she uses tonfa as a weapon.
Originally, she was created as a substitute heroine while Kenshin and Kaoru were separated due to the story's development. The story was heading in a somewhat dark direction, so she was drawn as brightly as possible, resulting in a character that is a mix of Kaoru and Yahiko. Her concept was based on a reader's suggestion for an "Oniwabanshū kunoichi," and she was designed not to have romantic feelings for Kenshin to avoid complicating the story further. Her design motif was often compared to Nakoruru by various people, but the author commented, "I thought she was closer to Mai Shiranui or Yuri Sakazaki from the same company..." She is a favorite character of the author and served as a motif for Kazuki Muto, the protagonist of the later serialized work "Buso Renkin."
In the complete edition volume 7, her hairstyle was changed to an outward flip, making her more boyish. She uses wedge-shaped shuriken attached to her gauntlets as weapons.
In the Kinema version, she appears in a single frame during a conversation between Saitō and Gein.
In the live-action film, she appears from the second movie "Kyoto Inferno." Her basic setting follows the original, but she wears less revealing clothing compared to the original and anime versions, and due to her Kyoto origin, she speaks with a Kyoto dialect. In battles, she mainly uses kicking techniques from the Oniwabanshū-style martial arts. In "Kyoto Inferno," she carries a dagger on her back but does not use it. In the fourth movie "The Final," during the final battle at Enishi's hideout, she leads the Oniwabanshū to support Kenshin in place of the severely injured Aoshi, who was injured protecting citizens from a bomb explosion in Tokyo. She introduces herself as "Oniwabanshū leader Shinomori Aoshi's proxy, Makimachi Misao, here!"

Kansatsu Tobikunai
Throws multiple kunai at once using the spring of her whole body.
Kansatsu Tobikunai: Kawasemi no Hashi
A "Kansatsu Tobikunai" technique aimed at underwater targets. In the story, it was used to destroy the "barrier" created by Kuroboshi's sea mines.
Keichō Geri
A flying kick said to be directly taught by Hannya. Depending on Misao's emotions at the time, words like "Angry -" or "Joyful -" are added to the name.
